Understanding Canine and Feline Infectious Disease Testing: Antigen vs Antibody

One of the most common sources of confusion in veterinary diagnostics is the difference between antigen and antibody tests. Both are essential tools, but they answer different clinical questions — and choosing the wrong one can lead to misinterpretation.

Antigen Tests: Detecting the Pathogen

Antigen tests detect the presence of the pathogen itself — a viral or bacterial protein — in the patient’s sample. Because they identify the infectious agent directly, they are the test of choice for diagnosing active infection.

Antigen tests answer the question: “Is the pathogen present right now?”

Common examples in veterinary medicine include antigen tests for canine parvovirus (CPV), feline leukemia virus (FeLV), and feline panleukopenia virus (FPV).

When to use antigen testing:

  • Confirming acute infection in a symptomatic patient
  • Screening new animals entering a shelter or cattery
  • Confirming a suspected outbreak

Antibody Tests: Detecting the Immune Response

Antibody tests detect the patient’s immune response to a pathogen — the antibodies produced after infection or vaccination. They are the test of choice for assessing immunity status.

Antibody tests answer the question: “Has the animal been exposed or vaccinated, and is it protected?”

Common examples include antibody tests for canine distemper (CDV), canine parvovirus (CPV), and feline calicivirus (FCV).

When to use antibody testing:

  • Confirming vaccine response
  • Determining whether a booster is needed
  • Assessing maternal antibody levels in puppies and kittens

The Value of Quantitative Results

Traditional rapid tests provide only a qualitative positive/negative answer. Quantitative fluorescence immunoassays go further by measuring the actual concentration of antigen or antibody. This enables:

  • Antibody titre monitoring — tracking whether protection is rising or waning over time
  • Objective treatment monitoring — confirming that antigen levels fall with successful therapy
  • Standardised reference ranges — reducing subjectivity in interpretation

Putting It Together

A well-designed diagnostic strategy uses both test types appropriately:

  1. Sick patient with compatible signs → antigen test to confirm active infection
  2. Vaccination check → antibody test to confirm protective immunity
  3. Monitoring therapy → serial antigen testing to track response
  4. Breeding or boarding screening → antibody panels to assess herd immunity
